Lipid metabolism in the development of cholesterol gallstones in hamsters. III. The effect of dietary cholesterol on its biliary concentration and lithogenesity.

چکیده
The effect of dietary cholesterol on the lithogenesity of bile in female hamsters was investigated. Biliary and serum lipids and serum lipoproteins of the hamsters fed with the normal diet supplemented with 1% cholesterol for 10, 20 and 30 days were analyzed. The synthesis rate of phospholipids and triglycerides were determined with (l-14C) palmitate incorporated into these lipids in the livers of the animals. The biliary cholesterol output slightly but significantly increased with a small and temporary increase in phospholipids 20 and 30 days after feeding the cholesterol diet. But these animals failed to show a significant change in the total and individual bile acids. Though the lithogenic index (Holzbach) gradually increased in accordance with the above change in the biliary lipid composition, it stayed far below the saturation point (1. 0) even 30 days after feeding the cholesterol diet, not causing the gallstone formation in the animal gallbladder. The striking increase in serum lipids including total cholesterol, phospholipids and triglycerides, and serum lipoproteins was, on the other hand, observed so shortly as 10 days after the cholesterol diet feeding. The cholesterol diet feeding also produced the increase in the synthesis of phospholipids and triglycerides contributing to the fatty acid composition of biliary phospholipids. These results suggest that dietary cholesterol brings about the increase in the biliary cholesterol output but that phospholipids synthesized in the liver maintain the lithogenesity far below the saturation point in the normal female hamsters. 141

Antilithiasic effect of beta-cyclodextrin in LPN hamster: comparison with cholestyramine.
Beta-Cyclodextrin (BCD), a cyclic oligosaccharide that binds cholesterol and bile acids in vitro, has been previously shown to be an effective plasma cholesterol lowering agent in hamsters and domestic pigs. This study examined the effects of BCD as compared with cholestyramine on cholesterol and bile acid metabolism in the LPN hamster model model for cholesterol gallstones. The incidence of ch...
متن کاملCholesterol, bile acid, and lipoprotein metabolism in two strains of hamster, one resistant, the other sensitive (LPN) to sucrose-induced cholelithiasis.
A comprehensive study of cholesterol, bile acid, and lipoprotein metabolism was undertaken in two strains of hamster that differed markedly in their response to a sucrose-rich/low fat diet. Under basal conditions, hamsters from the LPN strain differed from Janvier hamsters by a lower cholesterolemia, a higher postprandial insulinemia, a more active cholesterogenesis in both liver [3- to 4-fold ...
متن کاملEffects of Dietary Chitosan on Serum Lipid and Lipoprotein Concentration in Rats
Because the ingestion of some types of dietary fibers has been shown to influence on the lipid and lipoprotein levels, it is possible that chitosan influences on lipid metabolism. In the present study, the effects of chitosan on the serum, liver lipid and lipoprotein concentrations in rat were investigated. Serum lipid level in the treatment groups were decreased compared to that of the control...
متن کاملTHe influence of aging upon gallstones--changes in serum lipids, lithogenesis of bile and cross-sectional views of stones with age.
Differences in lithogenesity of bile, and the type of stones of the radiolucent gallstone patients were investigated by both the roentgenogram (solitary and multiple) and cross-sectional views in comparison between the patients of ages below 39 and above 40 either with or without hyperlipidemia and lipoproteinemia. Hypercholesterolemia was rare in patients of age below 39 in contrast to those o...
متن کاملDietary impact on biliary lipids and gallstones.
Although dietary factors influence bile lithogenicity and gallstone formation, the main dietary effect appears to be indirect, depending on an interaction between caloric consumption and gender-specific aspects of lipoprotein metabolism. Excessive energy intake elicits its detrimental effect by altering lipoprotein and hepatic cholesterol metabolism in association with hyperinsulinemia. Factors...
